How about something subtle? Write a home page announcement "You're invited! Join us for free food to launch our new FoE/AQIP project [or some other acronym that no one will understand] in room T425 [or some other room that doesn't exist, directing them to a bathroom or closet would work too.]" Sounds like you have a fun boss :)

We'd never get away with the April Fools joke... although I'd love to do it. The only thing that I can get away with is contra code on one website... but that is top secret :)

Anyone planning a joke for 2011? I'd love to do something funny that would make the rounds on Facebook. I've seen several schools do things to their home pages, but can't find any archived examples.

What about some joke/message hidden in the source code of the home page?  Perhaps a little ASCII art? That way the prank that won't screw with your conversions or freak out the administration, but you can still get a little viral mileage if the right people know about it.  :-)
