We just finished up a Twitter contest at the University of North Alabama (@north_alabama).
The idea was that our 500th follower would get a prize package (t-shirt, coffee mug, etc.). It worked great in getting us a bunch of new followers, but right about when we were going to cross the 500 mark we had someone create several 'fake' accounts and then followed us w/ their real account to be #500.
The nice thing was though, the next day Twitter had suspended all those (and his) accounts in suspicion of spam. This all happened over a weekend, so come Monday morning we had a real (and legit) 500th follower. We're still climbing too.
We might try another contest to get people to use a certain hashtag. Best use of the tag wins sort of thing. Not sure when yet.
